If my mail server connect must by proxy,what should I add scripts in the ganglia-alert.Thanks.

It depends on which metrics you're trying to track, which mail server you're using, etc. If you're simply looking for a way to alert on failure to connect, the right tool would be an alerting package like Nagios.
